Bowker ISBN and GS1
Have seen many discussions around this already, and want to bring it again as I am facing the same issue.
I tried with ISBN initially, then I was asked to apply for GTIN exception. Now when I applied to GTIN exception, I am being asked to go GS1 database route which does not exist. Why cann't Amazon lookup ISBNs from Bowker directly?

Well Actually Amazon does, and so can you and I. What is the ISBN number? Place the ISBN in the Google browser, and what do you see?
That is what Amazon sees.
Now with that said, you did not give enough explanation of your problem to get much more help than that.
Are you trying to list your own book? Books published today , and for many years, have both the ISBN and GS1 UPC associated with them. Why doesn't yours have a UPC?
